Linkage specific, non-hydrolyzable di-ubiquitin is resistant to the activity of enzymes (DUB's) that cleave the isopeptide linkage between adjacent ubiquitin molecules.  It can be used to investigate binding interactions between di-ubiquitin and proteins that contain elements such as ubiquitin-associated domains (UBA's) or ubiquitin-interacting motifs (UIM's).  This product may also be useful in exploring the role of unanchored poly-ubiquitin chains in some signaling pathways.

Non-hydrolyzable Ub2 (K48) chains coupled to beads at 1 mg/ml (58 nmol/ml)

Use & Storage

Use: 

Prepare resin by rinsing with 10 volumes of water to remove ethanol storage buffer.  Equilibriate resin by washing with 10 volumes of desired start buffer.  Binding and elution of material is dependent on individual experimental conditions and requirements.

Storage: 
Polyubiquitin-agarose can be re-used if properly maintained. After use, clean resin with a wash cycle of 5 volumes 100 mM HEPES pH 8.0, 500 mM NaCl followed by 5 volumes 100 mM NaOAc pH 4.5, 500 mM NaCl. Repeat twice, then rinse resin with a low salt buffer. Store resin at 4 degrees Celsius in neutral aqueous buffer containing 1mM NaN3 or 20% ethanol as a preservative. DO NOT FREEZE.
