Cell fraction containing the full HeLa cell compliment of E1, E2s, E3s, 20S and 26S proteasomes, isopeptidases, and PA28. Useful in assays to demonstrate ubiquitin conjugation and degradation.
Use & Storage
Use:
Ideal for demonstrating ubiquitin proteasome degradation/ conjugation of radiolabeled or immuno-detectable substrates. Typical assay concentration for extract is 0.5-4 mg/ml. Isopeptidase inhibitor (U-201) and a proteasome inhibitor are highly recommended for the efficient accumulation of ubiquitinprotein conjugates.
Storage:
Store at -80°C. For best results, aliquot and freeze after first set of experiments to avoid multiple freeze/thaw cycles. Literature
