Also known as: FII
Protein fraction from a cell extract that binds to an anion exchange resin. Essentially ubiquitin-free and ATP-free, FII contains E1, most E2s, some E3s, UCHs, and the proteasome (20S and 26S).
Use & Storage
Use:
Ideal for demonstrating ubiquitin- and ATP-dependent degradation/conjugation of radio-labeled or immuno-detectable substrates. The inclusion of an isopeptidase inhibitor and a proteasome inhibitor are highly recommended for the efficient accumulation of ubiquitin protein conjugates. Typical assay concentrations for FII are 0.5 - 4 mg/ml.
Storage:
Store at -80°C. For best results, aliquot and freeze after first set of experiments to avoid multiple freeze/thaw cycles. Literature
