Biochemically, UBE2K (E2-25K) can catalyze the formation of K48-linked ubiquitin chains in the absence of an E3 ligase. This enzyme has a unique C-terminal UBA domain that plays a critical role in E1 selectivity and chain synthesis. Recently, UBE2K (or E2-25K or HIP2) has been shown to bind to the N terminus of huntington and may play a role in Huntington disease.
