USP25 (Ubiquitin Specific Protease 25) is a deubiquitinating enzyme of the C19 family that is expressed in many human tissue types.  USP25 activity is modulated by a SUMOylation event that is dependent on the presence of a SUMO-interaction motif (SIM) located in the amino-terminal region of all USP25 isoforms.  SUMOylation occurs within one of the two Uibquitin interaction motifs (UIMs) that are required for efficient in vitro hydrolysis of poly-Ubiquitin chains by USP25.  While SUMOylation impairs USP25 activity against poly-Ubiquitin substrates, it does not affect its hydrolysis of Ubiquitin-AMC (U-550).  This protein contains a C-terminal 6-His tag.
