UBE1L2 Activating E1 Enzyme
(Uba6) is a newly identified E1 activating enzyme for ubiquitin. This enzyme charges a newly
identified E2 enzyme, Use1, which is not charged by UBE1 (Uba1). It also charges various E2 enzymes
including all UbcH5 isoforms, UbcH6, UbcH7, UbcH10 and UbcH13. Additionally, FAT10 forms a
thiolester complex with Use1 in the presence of UBE1L2 but not with UBE1. The UBE1L2 enzyme
does not activate other UBLs (including SUMO, NEDD8, ISG15, UFM1), indicating specificity for
ubiquitin and FAT10.

Complex Formation with various E2 enzymes in the presence of the

containing GST-Uba6 (50 nM), E2 enzymes (5μM) and Ubiquitin
(100μM) were incubated in the
absence or presence of Mg-ATP (5mM) at 37 degrees C for 60
minutes. Proteins were separated by 16% SDS-PAGE
under non-reducing conditions. The § symbol denotes the Ub-E2
thiolester complexes.

(5μM) and FAT10 (10μM) were incubated in the absence or presence
of Mg-ATP (5mM) at 37 degrees C for 60
minutes. Proteins were separated by 4-20% SDS-PAGE under
non-reducing conditions. Asterisks (*) denote the E2
enzyme proteins.
