p53 Ubiquitination Products

Boston Biochem now offers a comprehensive set of tools for the study of p53 regulation by ubiquitin. The stability of the tumor suppressor p53 is regulated via the ubiquitin-proteasome pathway (UPP). p53 is ubiquitinated by E3 ligases (including Hdm2, Pirh2, COP1, p300 and E6AP) and deubiquitinated by enzymes including USP7 (HAUSP) and USP8. Figure 1A. Ubiquitination of p53 by the Hdm2 E3 ligase. The substrate protein p53 (SP-450) was ubiquitinated with Hdm2 (E3-200) using the p53 Conjugation Kit (K-200). Western blotting with a p53-specific monoclonal antibody (A-200) was performed to detect the formation of mono- and poly-ubiquitinated protein.
Figure 1B. Deubiquitination of p53 by USP7 and USP8 enzymes. Poly-ubiquitinated p53 (SP-450) was then used as substrate for USP7 (E-518) and USP8 (E-520) and incubated at 37oC for 1 hour. Western blotting with a p53-specific monoclonal antibody (A-200) was performed. The lower panel shows a shorter exposure to indicate the appearance of p53 after ubiquitin chain removal.
